Methylglyoxal

Methylglyoxal is a 2-oxo aldehyde derived from propanal. It has a role as a human metabolite, a Saccharomyces cerevisiae metabolite, an Escherichia coli metabolite and a mouse metabolite.
When incubated with methylglyoxal (MG) alone, the insulin peak on the HPLC chromatogram weakened in a concentration-dependent manner with increasing MG concentration until it completely disappeared at 100 equivalents, as shown in Figure 1a. However, under the same conditions, incubation of insulin with MG in the presence of D-Dap-L-Leu (dipeptide/MG = 1.1 equivalents) significantly reduced this trend (Figure 1b).

Reagents and conditions: (a) KOH, EtOH, 80 °C, 90%; (b) oxalyl chloride, DCM, then NH4OH,; (c) Tf2O, TEA, DCM, two step 36%; (d) NBS, CCl4, benzoylperoxide, reflux, 97%; (e) NaN3, DMF, 85 °C (f) PPh3, DMF/H2O, 40 °C, 85% over two steps; (g) ethylsulf-onylethene, TEA, MeOH, 99%; (h) 4-fluoro-3-trifluoromethylphenyl-acetic acid, EDC, HOBt, NMM, DMF, rt, 61% over two steps. Enantiomerically pure products were isolated by chiral phase HPLC.

1,3-Dimethylcycloprop-1-ene

The monosubstituted product was reacted directly with p-cresol without purification to afford the desired phenylphosphonate 2a.
