What is the role of cyclodextrins in enhancing the solubility and dissolution of raloxifene?
Label:chem
Topic
Cyclodextrins are cyclic oligosaccharides with hydrophobic cavities and hydrophilic exteriors that can form inclusion complexes with poorly soluble drugs, thereby improving their aqueous solubility and bioavailability.
Answer
Cyclodextrins enhance the solubility and dissolution of raloxifene by forming stable inclusion complexes. The study found that hydroxypropyl-β-cyclodextrin (HP β-CD) formed the most stable complex with raloxifene, resulting in an 8-fold increase in solubility compared to the untreated drug. The coprecipitation method further improved the solubility and dissolution rates of raloxifene when combined with cyclodextrins.
