How does thidiazuron (TDZ) affect the in vitro shoot multiplication of Húsvéti Rozmaring apple scion?
Label:chem
Topic
Thidiazuron (TDZ) is a commonly used cytokinin known for its ability to stimulate shoot multiplication in plant tissue culture. However, its effects can vary depending on the plant species and genotype. The study investigates the impact of TDZ on the in vitro shoot multiplication and quality of Húsvéti Rozmaring apple scion.
Answer
TDZ was found to be the most efficient cytokinin for enhancing the number of shoots per explant, with the highest significant multiplication rate of 5.40 shoots/explant achieved at a concentration of 2 µM. However, TDZ resulted in stunted, dwarf shoots with pale green leaves, making it difficult to separate the shoots for further subculturing. Therefore, while TDZ is effective for shoot multiplication, it negatively impacts shoot quality.
