Dabigatran etexilate mesylate exhibits strong pH-dependent solubility, showing good solubility in strong acidic media and significantly reduced solubility above pH 3. In aqueous media, especially above pHmax (around pH 2.5 for dabigatran etexilate mesylate), the mesylate salt undergoes rapid disproportionation to the free base, which then precipitates as a tetrahydrate, leaving free methanesulfonic acid in solution. The equilibrium solubility and intrinsic dissolution rates are very similar for both anhydrous forms I and II.
